Summary / Zusammenfassung Light propagation in tissue is diffusive, i.e. light photons detected are scattered multiple times, the loss of coherence leading to the loss of the spatial information. As a consequence, the accurate determination of fluorophor concentrations is not feasible when using currently available commercial systems, which operate in the so-called reflectance imaging mode, The measured signal intensity critically depends on the length of the path the light has to travel through tissue: e.g. a diffuse weak signal may be due to a low intensity (low concentration) distributed light source close to the surface or to a high intensity focal light source embedded deep in tissue. Quantitative data analysis requires knowledge of the fluorophor distribution in three dimensions. The project aims at development of 3D diffuse optical tomography. The system should allow for contact free measurements, i.e. the detectors are not in contact with tissue, which is a prerequisite for imaging studies of the brain. This implies that image reconstruction algorithms have to consider optical effects at the tissue surface.
