Investigating Complex Folding and Misfolding Mechanisms with Multiparameter Fluorescence Spectroscopy

Summary / Zusammenfassung In this collaborative project we investigate the competition between protein folding and misfolding using components of biochemistry, spectroscopy and theoretical biophysics. We combine the powerful method of single molecule multiparameter fluorescence detection with resonance energy transfer between up to three dyes, advanced strategies for fluorophore attachment and sample immobilization, parallel two-color excitation, photon detection in up to six channels and the development of theoretical methods including molecular simulations for the structural interpretation of the results. This will not only contribute to understanding the causes of the potentially harmful process of protein misfolding, but also extend the available methods for the single molecule analysis of complex systems.
