Title / Titel Physiological and molecular characterization of multispecific organic anion transporting polypetides

Summary / Zusammenfassung Organic anion transporting polypeptides (rodents: Oatps; human: OATPs) form an important superfamily of solute carriers with a wide spectrum of amphipathic transport substrates. Many members have a broad and overlapping substrate specificity and are expressed in a variety of different tissues. With their ability to transport endo- and xenobiotics and their predominant expression in organs important for excretion of potentially toxic compounds they appear to be part of the overall body detoxification mechanisms. However, the toxicological and physiological significance of certain superfamily members remains to be determined. Furthermore, the molecular basis of the broad substrate specificity is only poorly understood.
To understand the toxicological and physiological importance of organic anion transporting polypeptides and the molecular basis of their broad substrate specificity a detailed functional and molecular characterization is required which includes 1) further characterization of substrate transport mediated by Oatps/OATPs, 2) identification of the substrate binding site(s) of Oatps/OATPs 3)purification of functionally active protein and 4) the cloning and characterization of additional members of the Oatp/OATP superfamily.
