Title / Titel Effect of Interferon-alpha on the HIV-1 Latent Reservoir in HIV-1 / HCV coinfected Patients

Summary / Zusammenfassung Background: One major barrier to eradicate HIV-1 is the latent reservoir already formed during acute infection. It has been demonstrated that administration of pegylated interferon alpha (IFNα) in HIV-1 infected individuals led to a reduction of the latent reservoir.

Study Aims: To investigate the effect of standard of care (SOC) Hepatitis C treatment consisting of IFNα and ribavirin (RBV) on the HIV-1 latent reservoir in HIV-1/HCV coinfected patients from the SHCS.

Study Design: Retrospective analysis of the HIV-1 latent reservoir using blood samples from HIV-1/HCV coinfected patients with a history of SOC Hepatitis C by estimating decay kinetics of the HIV-1 latent reservoir. Only patients with stably suppressed HIV-1 RNA will be included. Proviral DNA and different HIV-1 RNA forms will be quantified in peripheral blood mononuclear cells (PBMC) of a variety of clinically meaningful time points (i.e., 6 months before INFα/RBV administration, immediately before first administration of INFα/RBV, at months 6 and 12 during INFα/RBV treatment, and then annually during follow-up). Cases will be compared to matched control groups (HIV-suppressed/no IFN Trt).
