Title / Titel The epigenome of colorectal transformation: from early tumors to liver metastases.

Summary / Zusammenfassung The epigenetic changes that characterize the different stages of neoplastic transformation of colorectal epithelial cells are so numerous that high-throughput analytical methodologies are the only way to identify them all and evaluate their relevance to tumor progression and spread. We also believe that an integrated approach to the study of the different determinants that shape the epigenetic landscape over the whole genome of colorectal neoplasms (i.e. epigenome) will enable us to decipher the molecular mechanisms underlying the malignant cell phenotype in these tumors and their drug resistance.
We will exploit the enormous potential of next-generation sequencing for comprehensive detection of the epigenetic alterations characterizing different stages of colorectal neoplasia. Pinpointing the changes that drive the tumor from one stage to the next can be useful for several reasons. Crucial, early alterations can be targeted to improve diagnostic and prognostic aspects of colon-cancer management. Those identified in primary cancers, liver metastases, and putative cancer stem cells can be exploited to develop rational, targeted strategies for dealing with advanced disease (biomarkers of progressive and relapsing disease, indicators of response to drugs, etc.).
