Summary / Zusammenfassung One of the current research projects focuses on the improvement of the detection of gene methylation status in fresh and formalin-fixed tissues. Special emphasis is set on improving reproducibility and quantitative aspects in order to fulfill the strict quality requirements for diagnostic applications.
Furthermore, we are working on improving the detection of specific translocations using different PCR-techniques. Special fields of interest are lymphomas and soft tissue tumor diagnostics on fresh and formalin fixed-tissues.
Finally, we regularly establish sensitive gene mutation analyses required for prognosis or for prediction of target therapies in cancer.

Our routine diagnostics in the laboratory of molecular diagnostics currently includes:
- Clonality assessment of lymphoproliferative diseases with Southern blot and PCR
- Detection of t(14;18), t(11;14) and t(11;18) translocations in lymphoma with standard, RT- and long-distance PCR
- Detection of t(X;18) in synovial sarcoma, t(11;22) and t(21;22) in Ewing sarcoma, t(12;16) and t(12;22) in myxoid liposarcoma and t(12;22) in clear cell sarcoma in fresh-frozen and formalin-fixed tissues using RT-PCR
- Detection of loss of heterozygosity (LOH) of specific microsatellite-markers in bioptic and cytological samples of bladder carcinoma patients using PCR and capillary electrophoresis
- Detection of loss of heterozygosity (LOH) of specific microsatellite-markers for the diagnosis and prognosis of oligodendroglioma
- Detection of microsatellite instability in colon carcinoma
- Gene mutation analysis (EGFR, ERBB2, KIT, PDGFRA, KRAS, BRAF, CEBPA, CTNNB1, GNAS, HFE, PRNP, JAK2, IDH1 and 2) by PCR and direct DNA sequencing
- Detection of mycobacterium tuberculosis infections in formalin-fixed tissues with PCR
- Detection and typing of non-tuberculosis mycobacteria in formalin-fixed tissues with PCR and sequencing
- Detection and typing of human papillomavirus infections from cytological preparations and biopsies using PCR and DNA sequencing
- Detection of human herpesvirus-8 with PCR
