| Summary / Zusammenfassung | Ribavirin in combination with pegylated interferon is the current standard treatment for chronic hepatitis C in HIV/HCV coinfected individuals. Optimal ribavirin exposure is important for a successful anti-HCV treatment. The correlation between ribavirin dose and ribavirin plasma concentration level in HIV/HCV coinfected patients is less well documented than in HCV monoinfected patients; indeed, little data exist about the influence of HIV-associated factors on ribavirin pharmacokinetics. Until now, in HIV/HCV coinfected patients study results are conflicting whether ribavirin therapeutic drug monitoring correlates with virological response. The utility of ribavirin therapeutic drug monitoring in this population is not determined yet. The study aims are first, to assess ribavirin plasma concentrations as a function of dosage; secondly, to identify factors influencing ribavirin concentration levels and third, to evaluate a correlation between ribavirin concentrations at steady-state and early and sustained virological response. In this retrospective analysis we will assess ribavirin concentration levels in stored plasma samples in 396 Swiss HIV Cohort Study (SHCS) participants with HIV/HCV coinfection on ribavirin and pegylated interferon treatment. In addition to the SHCS database a chart review will provide HCV specific patient information. In univariable and multivariable generalized linear models we will identify predictors for ribavirin concentration levels. With logistic regression analyses we will evaluate the association between ribavirin plasma levels and early and sustained virological response. |
