Title / Titel The effect of sertindole on sensory gating and cognition in healthy humans: An ERP study

Summary / Zusammenfassung Recent data suggest that healthy human subjects with low sensory gating levels may provide a useful translational model to investigate pharmacological manipultions on gating functions and their association to cognitive performance. Specifically, we and others found that the atypical antipsychotics clozapine increases PPI in clinically unaffected healthy subjects with low baseline PPI (Vollenweider et al., 2006;Swerdlow et al., 2006), while typical antipsychotics such as haloperidol did not improve PPI in healthy subjects with low PPI levels (Csomor et al., 2007). In accordance with Bitsios and co-workers we found also that healthy humans with low PPI levels show impaired spatial working memory performance (Giakoumaki et al., 2006;Bitsios et al., 2006). These findings may implicate a role for the 5-HT2-R in cognitive performance. Moreover impairments in working memory in healthy human volunteers have also been linked to allelic variants in the 5-HT2A receptor (de Quervain et al., 2003).

This study aim at investigatin whether the novel 5-H2/6 and D2 antagonsist sertindole will increase PPI and P50 supression in subjects with low PPI levels associated with alterations in cognitive performance. Various domains of cognitve erformance shall be investigated using the CANTAB test battery.
