| Summary / Zusammenfassung | Although protein folding is one of the most fundamental processes in biology, our understanding of the underlying mechanisms is still far from complete, and hypotheses arising from recent theoretical considerations have proven difficult to test experimentally. Work from our laboratory and others has shown that single molecule fluorescence spectroscopy can be used to quantitatively address some of the central questions that have eluded investigation with classical ensemble techniques, such as the structure and dynamics of unfolded proteins at near-physiological conditions. Here we use of single molecule Förster resonance energy transfer (FRET) to study the heterogeneous structure and dynamics of proteins in several different non-native states with direct relevance for cellular conditions. | ||
