Summary / Zusammenfassung Recently, it has become evident that the Roche Taqman assay might have a problem to accurately determine HIV-plasma RNA levels in some HIV-1 infected individuals. In Zurich, we have two unambiguously documented cases, in which the Taqman assay failed to measure HIV RNA accurately. The problem seems to be particularly but not exclusively prominent in non-B subtypes.
Study Aims
The aim of this study is to propose an approach to address this potentially serious problem in the SHCS and to obtain an estimate for the magnitude of HIV RNA underdetection by the Taqman assay. To have accurate HIV-RNA values is of utmost importance for 1) individual patient management, 2) for epidemiological analysis of SHCS projects, particularly for studies evaluating patients with subtype non-B. Furthermore, this study will help to assess further strategies and in particular, whether the Roche Taqman assay may still be a suitable assay for HIV-1 RNA-measurement for rare HIV-1 subtypes.
