Title / Titel Vitamin B12 for targeted cytotoxic drug delivery

Summary / Zusammenfassung Vitamin B12 is an essential biomolecule. It is used in large amounts by rapidly proliferating cells or by bacterial infections. These reasons makes it a favoured targeting molecule for diagnosis or therapy of cancer or bacterial infections. In recent projects, we have shown that B12 can be used for carrying cisplatin like compounds into cancer cell or bacteria. The cytotoxic cisplatin compounds were bound in a unpredecented way to B12, namely by direct coordination to cyanide in B12.
In this project, we will unambigously establish that cisplatin like compounds delivered by B12 targeting can have a cytotoxic effect on rapidly proliferating cells. Furthermore, organic cytotoxic or antibacterial molecules will be bound to B12 via coordination to Pt(II). This allows not only to bring platinum into cells but also organic cytotoic agents.
Since Pt(II) complexes are not the only metal-based drugs which are biologically active, we will attempt to incorparate other metal complexes with Au, Hg, Ru or Rh into cells. Metal complexes based on these elements are well known cytotoxic agents and can replace / complement Pt.
