Title / Titel In vivo Correlates of HIV-1 Latency During Antiretroviral Therapy; Individualized Approaches to Quantify Highly Variable Viral Nucleic Acids

Summary / Zusammenfassung The latent form of HIV prevailing during antiretroviral therapy is the main impediment that forestalls eradication of HIV or long-lasting remission of the infection after cessation of therapy. Recently evidence has come forth that HIV-DNA may reflect the totality of different forms of latently infected cells in vivo. Furthermore, latently infected cells have been shown to display viral transcriptional patterns significantly different from virus producing cells. To monitor kinetics of thelatent and virus producing reservoirs during combination antiretroviral therapy (cART), we have developed a novel highly reliable algorithm for HIV-nucleic acid quantification by combining powerful bioinformatics with state of the art PCR technology. In the current study we aim to validate these method and to apply it to a cohort of 50 acutely infected patients to identify correlates of virus control and immunological benefit of early antiretroviral therapy.
